成果简介NOVELTY - A primer pair comprising a 33 base pair sequence (SEQ ID NO: Not defined) fully defined in the specification and comprising a 35 base pair sequence (SEQ ID NO: Not defined) fully defined in the specification for introducing a mutation into the nucleotide sequence encoding the potato translation initiation factor eIF4E2 using site-directed mutagenesis, resulting in the CCC triplet being replaced by a CTC triplet, leading to change in the amino acid at position 59 from proline to serine, is new. USE - The primer pair is useful for introducing mutation into nucleotide sequence for producing a functional translation initiation factor eIF4E2 of potato with a p59s mutation. ADVANTAGE - The primer pair produces functional translation initiation factor eIF4E2 of potato (Solanum tuberosum) with a p59s mutation that disrupts its interaction with a wide spectrum of viral genome-linked protein (VPg) variants of potato virus Y. DETAILED DESCRIPTION - An INDEPENDENT CLAIM is included for a nucleic acid molecule, which encodes a mutant translation initiation factor eIF4E2 of potato with a proline to serine substitution in the amino acid sequence fragment comprising an 23 amino acid sequence (SEQ ID NO: Not defined) fully defined in the specification or a sequence exhibiting at least 80% identity to the above sequence in any variety of cultivated potato or other plant.
