成果简介NOVELTY - Method for isolating nucleic acids from sectional material involves performing mechanical processing of the biomaterial, treating the biomaterial sample with a 400 μ l denaturing buffer solution comprising 0.1 M tris-hydrochloric acid, 0.1 M EDTA, 0.5 M sodium chloride, 1% sodium dodecyl sulfate (SDS) and 20 μ l proteinase K solution at a concentration of 20 mg/ml, incubating the mixture for 5-6 hours and precipitating non-target biopolymers and cellular debris by centrifugation in a 5 M sodium chloride saline solution with a volume of 500 μ l at a speed of 10000-15000 g, taking an aliquot of 400 μ l from the upper layer and transferring to a new tube, adding 400 μ l isopropyl alcohol to the collected supernatant, vortexing and centrifuging for 10 minutes at 10000 g, removing the supernatant, washing the sediment twice and centrifuging with 75% ethanol, dissolving the sediment containing nucleic acids in deionized water, and storing at -20° C. USE - The method is useful for isolating nucleic acids from sectional material.
