| 成果简介 | NOVELTY - Polymeric nano-formulation comprises nano-particles, where each particle is composed of component 1 and a component 2 in a ratio selected from the range of 1:3-3:1. The component 1 comprises temozolomide hydrazide (TMZ-H) conjugated to an amphiphilic polymer comprising a polycarbonate block and a polyethylene glycol block. The component 2 comprises an amphiphilic polymer comprising a polyester or polycarbonate block and a polyethylene glycol block. USE - Polymeric nano-formulation for delivery of temozolomide. ADVANTAGE - The nano-formulation has higher drug loading capacity and improved stability. DETAILED DESCRIPTION - INDEPENDENT CLAIMS are included for: 1. a pharmaceutical composition, which comprises the polymeric nano-formulation of temozolomide; and 2. a method for preparing the polymeric nano-formulation of temozolomide, which involves: (a) dissolving component 1 and component 2 in dichloromethane: ethanol of ratio 3:1 and subjecting the mixture to the thin film hydration process resulting in the formation of a thin film of a hybrid composition of components 1 and 2; (b) purging the thin film with dried nitrogen gas for 30-60 minutes and redispersing the thin film using a dispersive medium being phosphate-buffered saline to yield a dispersion mixture; and (c) probe-sonicating the dispersion mixture for 15-30 seconds at 20% amplitude under ice-cold conditions, followed by centrifugation at 10,000-15,000 revolutions per minute (rpm) for 15 5-15 minutes, resulting in a supernatant comprising the nano-particle formulation of temozolomide. |
