| 成果简介 | NOVELTY - Folate conjugated carfilzomib loaded lactoferrin nanoparticles formulation is prepared by: (a) preparing carfilzomib loaded lactoferrin nanoparticles (CFZ-LFNPs), (b) conjugating folic acid to the nanoparticles (CFZ-LF-NPs) obtained to obtain folate functionalized carfilzomib loaded lactoferrin nanoparticles (FA-CFZ-LF-NPs), and (c) performing lyophilization of folate functionalized carfilzomib loaded nanoparticles. USE - Folate conjugated carfilzomib loaded lactoferrin nanoparticles formulation for treating ovarian cancer. No biological data given. ADVANTAGE - The folate conjugated carfilzomib loaded lactoferrin nanoparticles formulation reduces the frequent dosing and enhance the prolongation in fatal and asymptomatic disease like ovarian cancer, thus enhancing the circulation and retention. DETAILED DESCRIPTION - INDEPENDENT CLAIMS are included for: 1. a method for preparing carfilzomib loaded lactoferrin nanoparticles (CFZLF-NPs), comprising: (a1) dissolving 0.25-2.0 wt. vol.% lactoferrin in 0.1-1.0 vol.% solution of ethanol-water at a pH of 5.0 to obtain a clear solution, dissolving the 0.25 wt. vol.% drug carfilzomib in 0.1-1.0 vol.% ethanol-water at a pH of 5.0, mixing both the solutions with stirring, and desolvating the nanoparticles by adding a mixture of ethanol-water at ratio of 2:1 and raising the pH of the solution to 6.0 by addition of 0.1 M phosphate buffer at 30-40℃; (b1) further adding 0.05 -1.0 vol.% glutardehyde into the turbid mixture and stirring for 12 hours to obtain suspended nanoparticles which are separated by centrifugation of the suspension at 5000 rpm and purifying the nanoparticles by washing with deionized water to obtain carfilzomib loaded lactoferrin nanoparticles (CFZ-LF-NPs); 2. a method for preparing the folate-conjugated carfilzomib loaded lactoferrin nanoparticles (FA-CFZ-LF-NPs), comprising: (a2) reacting activated folate i.e. folic acid-N-hydroxysuccinimide ester (FA-NHS) with the carfilzomib loaded lactoferrin nanoparticles (CFZ-LF-NPs) in equal molar proportions in 0.2 M sodium carbonate-sodium bicarbonate buffer of pH 10.0, for 8 hours with stirring at room temperature; and (b2) purifying the product FA-CFZ-LF-NPs by dialysis using the same carbonate-bicarbonate buffer to obtain the desired folate conjugated carfilzomib loaded lactoferrin nanoparticles (FA-CFZ-LF-NPs) product; and 3. a method for performing lyophilisation of folate-conjugated carfilzomib loaded lactoferrin nanoparticles (FA-CFZ-LF-NPs), comprising: (a3) mixing the nanoparticles (FA-CFZ-LF-NPs) with mannitol in a weight ratio of 1:2 in deionized water and storing the solution at 4℃ for 24 hours; (b3) lyophilizing the mixture at -25℃ for 24 hours in the first cycle for primary drying followed by secondary drying at 25℃ at 200 mT vacuum for 8-10 hours offering a lyophilized powder with particle size of 80-90 nm and water contents less than 0.5 %. |
