DETAILED DESCRIPTION - INDEPENDENT CLAIMS are included for: Kit for in-vitro development of preantral follicles and release of oocytes, which comprises first container comprising lyophilized stromal cell secretomes, second container comprising Dulbecco's modified eagle medium (DMEM), third container comprising one supplement, and a leaflet providing use instruction, where the third container comprises a separate compartment for each of supplements selected from insulin-transferrin-selenium (ITS), epidermal growth factor (EGF), non-essential amino acids (NEAA), bovine serum albumin (BSA), recombinant follicle-stimulating hormone (rFSH), fetal bovine serum (FBS) and human chorionic gonadotrophin (hCG); and Method for in-vitro development of preantral follicles and release of oocytes, which involves: (1) providing human mesenchymal stromal cells from bone marrow (BM), adipose tissue (AT), and Wharton's jelly (WJ); (2) cryopreserving said human mesenchymal stromal cells using a freezing mixture to obtain cryopreserved human mesenchymal stromal cells; (3) subjecting the cryopreserved human mesenchymal stromal cells to thawing and culturing till 70 to 80% confluency in a medium comprising Dulbecco's modified eagle medium (DMEM), followed by centrifugation and filtering to obtain stromal cell secretomes; (4) lyophilizing stem cell secretomes using 4 wt.% Trehalose to obtain lyophilized stem cell secretomes; and (5) culturing pre-antral follicles in a Dulbecco's modified eagle medium (DMEM) enriched with the lyophilized stem cell secretomes and supplementing with bovine serum albumin (BSA), insulin-transferrin-selenium (ITS), epidermal growth factor (EGF), non-essential amino acids (NEAA), recombinant follicle-stimulating hormone (rFSH) and fetal bovine serum (FBS), where the supplementing is carried out in every alternate day for 8-12 days and the recombinant follicle-stimulating hormone (rFSH) is replaced by human chorionic gonadotrophin (hCG) from day 5, where the lyophilized stromal cell secretomes are mixture of bone marrow mesenchymal stromal cell secretome, Wharton's jelly mesenchymal stromal cell secretome, and adipose tissue mesenchymal stromal cell secretome. |
