成果简介NOVELTY - An engineered antimicrobial peptide (AMP) SR17 comprising a short (less than 20 amino acids), and synthetic acyclic peptide under 2 kDa, end-capped at both N- and C-terminus having specified sequence (SEQ ID NO: 1), is new. USE - Engineered antimicrobial peptide for killing gram-positive bacteria and gram-negative bacteria such as Escherichia coli, Pseudomonas aeruginosa, Acinetobacter baumannii, and Staphylococcus aureus. Test details are described but no results given. ADVANTAGE - The engineered AMP has negligible toxicity towards mammalian cells at 0.2-150 µm and insignificant hemolysis of human erythrocytes over 0.2-150 µm (0.55-284 µg/ml), and broad-spectrum antimicrobial activity with multimodal mechanism of action and minimum inhibitory concentration of 4.6-37.5 µm (8.8-71 µg/ml) preferably against Escherichia coli, Pseudomonas aeruginosa, Acinetobacter baumannii, and Staphylococcus aureus bacterial strains when incubated for over 18 hours, improved thermal and protease stability, membrane permeability, membrane damage, and DNA binding, and engineered AMP is easily tunable, and is resistant to proteolysis when incubated for ~4 hours with trypsin DETAILED DESCRIPTION - An engineered antimicrobial peptide (AMP) SR17 comprising a short (less than 20 amino acids), and synthetic acyclic peptide under 2 kDa, end-capped at both N- and C-terminus having CH3CO-(D-Pro)-Val-Thr-Aic-Leu-Lys-Aib-Gly-(D-Arg)-Lys-Ile-(D-Pro)- Gly-Trp-Arg-(D-Thr)-NH2(SEQ ID NO: 1), is new, where D represents D-chiral amino acid.
