成果简介NOVELTY - Use of an antibody (A1) that specifically binds to the variable domain of the β chain of a human T-cell receptor of the T cell receptor β variable 5-1 (TRBV5-1) family for preventing or treating type 1 diabetes mellitus in a subject, where the complementarity determining region (CDR) of this variable domain has an amino acid sequence substantially similar to an amino acid sequence comprising SEQ ID NO: 21-35 (sequences not defined). USE - The antibody is useful for preventing or treating type 1 diabetes mellitus in subject, where the subject has at least one autoantibody comprising an antibody to insulin (IAA), an antibody to glutamate decarboxylase (GAD), an antibody to islet cells (ICA), an antibody to tyrosine phosphatase (IA-2), or an antibody to the zinc transporter (ZnT8), and the subject has been found to have at least one T-lymphocyte clone that contains the variable domain of the β chain of a human T-cell receptor of the TRBV5 family (all claimed). No biological data given. ADVANTAGE - The antibody: does not substantially bind to variable domains of a human T-cell receptor that do not belong to the TRBV5-1 family; binds to the TRBV5-1 segment of the variable domain of the β chain of the human T-cell receptor with a KD 10 nM; and results in the elimination of at least 90% of pathological T-lymphocyte clones that have a T-cell receptor comprising the CDR3 region of the variable domain of the β chain of the TRBV5-1 family. DETAILED DESCRIPTION - INDEPENDENT CLAIMS are also included for: preventing or treating type 1 diabetes mellitus, comprising administering the antibody (A1) to a subject; and a monoclonal antibody (A2) that specifically binds to the variable domain of the β chain of the human T-cell receptor of the TRBV5-1 family.
