| 成果简介 | NOVELTY - Rapid and sensitive method for detecting Entamoeba histolyticain human liver pus cells comprises: (i) extracting DNA from liver pus cell; (ii) performing loop-mediated isothermal amplification (LAMP) reaction with 25 µl reaction mix comprising 1.5µl of the mixture of 6 primers, 15 µl isothermal reaction mix, 1 µl sample DNA and reaction volume made with distilled water; and (iii) detecting Entamoeba histolyticaDNA. The primer/polynucleotide set comprises SEQ ID No. 1, SEQ ID No.2, sequence comprising 42 base pairs (SEQ ID NO: 3, 4), given in the specification, SEQ ID No.5 and SEQ ID No. 6. USE - Rapid and sensitive method for detecting Entamoeba histolyticain human liver pus cells. ADVANTAGE - The method is rapid, cost-effective and sensitive method for detecting amoebic liver abscess; provides 99% accuracy in detection of amoebic liver abscess in large number of samples simultaneously; provides accuracy in the results in lesser quantity of the primers and chemicals. DETAILED DESCRIPTION - Rapid and sensitive method for detecting Entamoeba histolyticain human liver pus cells comprises: (i) extracting DNA from liver pus cell; (ii) performing loop-mediated isothermal amplification (LAMP) reaction with 25 µl reaction mix comprising 1.5µl of the mixture of 6 primers, 15 µl isothermal reaction mix, 1 µl sample DNA and reaction volume made with distilled water; and (iii) detecting Entamoeba histolyticaDNA. The primer/polynucleotide set comprises SEQ ID No. 1 (catgggacaatgctgagg), SEQ ID No.2 (aatccttggttgacatcgtt), sequence comprising 42 base pairs (SEQ ID NO: 3, 4), given in the specification, SEQ ID No.5 (ggattttcacctctcccgtt) and SEQ ID No. 6 (agaacgaaagttaggggatcg). INDEPENDENT CLAIMS are included for: 1. a primer mixture comprising: forward inner primer (FIP, SEQ ID No.3) and backward inner primer (BIP, SEQ ID No.4) at a concentration of 40 pmol to 10 µl; loop Forward primer (SEQ ID No. 5) and Loop reverse primer (SEQ ID No. 6) at a concentration of 20 pmol - 5 µl; forward outer primer (F3, SEQ ID No.1) and backward outer primer (B3, SEQ ID No.2) each at concentration of 10 pmol to 2.5 µl; and 15 µl distilled water; and 2. a kit comprising the primer set; a negative control-a sample not containing nucleic acids; and positive control-nucleic acids extracted from detection target. |
