成果简介NOVELTY - Anti-cancer and immunomodulatory composition prepared from root biomass of Simarouba glauca, comprises 9-15 wt.% saponin, 7.5-11 wt.% carbohydrate, 6-8 wt.% alkaloids, 1.5-5.5 wt.% protein, and metal ions selected from iron, copper, and zinc and/or a combination, sodium, and potassium. USE - Anti-cancer and immunomodulatory composition prepared from root biomass of Simarouba glauca forinducing apoptosis human cancer cells. ADVANTAGE - The anti-cancer and immunomodulatory composition increases cytotoxic ability of immune cell towards cancer targets, exhibits significant in vivo anticancer activity against the four human cancer xenograft mice models. DETAILED DESCRIPTION - INDEPENDENT CLAIMS are included for: 1. a method for preparing an anti-cancer and immunomodulatory composition from root biomass of Simarouba glauca, comprising: (a) powdering the dried waste root biomass of Simarouba glaucausing mechanical grinder to obtain a root biomass powder having size of 0.9-10 mm; (b) adding aqueous methanolic solvent in volume ratio of 20: 80 Water: methanol in a weight ratio of 1:5-1:15 to the powder obtained in step (a) followed by filtration to separate the filtrate as crude aqueous methanolic extract and residual root biomass; (c) evaporating the crude aqueous methanolic extract on rotatory evaporator to separate the methanol; and (d) lyophilizing methanol free crude aqueous methanolic extract to obtain the solid crude aqueous methanolic extract, where the methanolic extract comprises 9-15 wt.% saponin, 7.5-11 wt.% carbohydrate, 6-8 wt.% alkaloids, 1.5-5.5 wt.% protein, and metal ions selected from 10 ppm iron, 5 ppm copper, and 2 ppm zinc, 20 ppm sodium, and 10 ppm potassium; and 2. an integrated method for treating root biomass of Simarouba glauca, comprising: (a1) powdering the dried waste root biomass of Simarouba glaucausing mechanical grinder to obtain a root biomass powder having size of 0.9-10 mm; (b1) adding aqueous methanolic solvent in volume ratio of 20: 80 water: methanol in a weight ratio of 1:5-1:15 to the powder followed by filtration to separate the filtrate as crude aqueous methanolic extract and residual root biomass; (c1) evaporating the crude aqueous methanolic extract on rotatory evaporator to separate the methanol; (d1) lyophilizing methanol free crude aqueous methanolic extract to obtain the solid crude aqueous methanolic extract; (e1) bleaching the residual root biomass by soaking the residual root biomass in acetate buffer of pH 4.5-5.0 containing 2.0-4.5 wt.% sodium chlorite (NaClO2) for bleaching at 50-70℃ for 2-4 hours; (f1) neutralizing the bleached biomass with water and treating with 0.5-1.5 M sodium hydroxide (NaOH) solution at 50-70℃ for 11-15 hours followed by washing with water till neutrality; (g1) suspending the washed biomass in 3-7% hydrocholoric acid (HCl) solution for 10-14 hours and washing the slurry with water till neutrality followed by lyophilization to get cellulose; and (h1) hydrolyzing the cellulose in 20-50% sulfuric acid for 30-90 minutes followed and lyophilizing after removing excess acid to obtain nanocellulose.
