成果简介NOVELTY - Carrying out semi-continuous cultivation for enhanced biomass and lipid production of Chlorella species BRE5, which involves: (a) cultivating Chlorella species BRE5 in the sterile blue green 11 (BG11) medium under controlled conditions, including a pH of approximately 7.1, temperature of 25 ± 2℃, light intensity of 167μmol/m/second, and aeration with carbon dioxide (CO2) at a rate of 0.25 vvm; (b) initiating cultivation in batch mode for 7 days to accumulate biomass without replenishing the culture medium; (c) transitioning to a semi-continuous mode of cultivation by periodically removing and replacing a portion of the culture medium with fresh BG11 medium to maintain the initial culture concentration. USE - Method for carrying out semi-continuous cultivation for enhanced biomass and lipid production of Chlorella species BRE5. ADVANTAGE - The harvested biomass is analyzed at the end of the cultivation period for total biomass yield, lipid yield, and fatty acid methyl ester (FAME) content to assess the effectiveness of the semi-continuous cultivation strategy. The replaced culture of Chlorella sorokiniana BRE5 strain with RP of both 2 days and 3 days with RR of 25% showed an elevated trend in biomass yield. The maximum yield in total biomass and total lipid of 7.27 g/L and 1.66 g/L, respectively, is observed with SRP9 (9th day as SRP) at RP of 2d and RR of 25%. The semi-continuous cultivation with SRP9 shows 4.35 and 3.69 times higher in total biomass and lipid yield. The biomass showsn fuel characteristics such as high heating value, better oxidative stability, and cetane number, which met international standards indicating its potential as a biodiesel feedstock. The optimization of SRP in semicontinuous cultivation under optimum RR and RP is a suitable strategy for biomass and biodiesel. DETAILED DESCRIPTION - Carrying out semi-continuous cultivation for enhanced biomass and lipid production of Chlorella species BRE5, which involves: (a) cultivating Chlorella species BRE5 in the sterile blue green 11 (BG11) medium under controlled conditions, including a pH of approximately 7.1, temperature of 25 ± 2℃, light intensity of 167μmol/m/second, and aeration with carbon dioxide (CO2) at a rate of 0.25 vvm; (b) initiating cultivation in batch mode for 7 days to accumulate biomass without replenishing the culture medium; (c) transitioning to a semi-continuous mode of cultivation by periodically removing and replacing a portion of the culture medium with fresh BG11 medium to maintain the initial culture concentration; (d) Conducting the semi-continuous cultivation at renewal rates (RR) of 25%, 50%, and 75%, and renewal periods (RP) of 1 day (24 hours), 2 days (48 hours), and 3 days (72 hours); (e) determining the optimal RR as 25% and the optimal RP as 2 days for maximizing biomass and lipid production; and (f) implementing the semi-continuous cultivation with the optimized RR and RP for a total cultivation period of 13 days.
