| 成果简介 | NOVELTY - Invention relates to a method of preparing a regenerating solution containing spider web proteins spidroin, fibroin, sericin. Method for preparing a regenerating solution containing spider web proteins spidroin, fibroin, sericin, including drawing and milling of silkworm cocoons, characterized by that it is additionally purified from sericin with 8-molar urea solution with temperature of 60 °C for 30 minutes to obtain a solution of sericin in urea, then silk is washed, silk is dried at temperature of 60 °C for 3 hours or at room temperature for 10 hours, the next step is to prepare solution 1:2:8 CaCl2:EtOH:H2O and the dried silk is dissolved therein for 2 hours at 75 °C, then the solution is centrifuged to precipitate undissolved particles, the solution is dialyzed in 20-fold volume of water at +4 °C, wherein the obtained sericin and urea solution is subjected to dialysis, the web is cut into pieces sized 1–2 mm and dissolved in concentrated trifluoroacetic acid (TFA) to obtain spidroin protein, further, the fluorinated web for cleaning is subjected to dialysis against water, the purified solutions of fibroin, sericin, spidroin are dried in a lyophilic drier, obtained proteins are mixed in a ratio of spidroin: fibroin: sericin 0.05:0.25:1 and diluted with distilled water, wherein beta-cyclodextrin is added for stabilization, in ratio of 1:1, as a preservative, sodium benzoate is used, which is 1/100 of the weight of the protein, and as a stabilizer, hyaluronic acid is added in amount of 2% of the total weight of the solution, wherein the final solution has the following composition: spidroin protein 0.05%, fibroin protein 0.25%, sericin protein 1%, beta-cyclodextrin 1.3%, sodium benzoate 0.013%, hyaluronic acid 2%, water 95.387%. USE - Biomedical materials technique. ADVANTAGE - Invention enables to obtain a regenerating solution containing fibroin, spidroin and sericin, to combine the properties of spider and silkworm silk in one solution, to increase their combined regenerative potential, to reduce formation of keloids, to accelerate regeneration of integumentary tissue, to reduce cosmetic defect of tissues in case of deep damages. 1 cl, 17 dwg |
