| 成果简介 | NOVELTY - Synthesizing biocompatible bisphosphonate (I), involves (a) mixing adipic acid ester derivative, phosphorous acid, and a halo phosphorous compound under stirring at 200-400 rpm to obtain a reaction mixture; (b) heating the reaction mixture within an hour at 60-100 °C under nitrogen atmosphere and refluxing for 14-16 hours followed by addition of cold water to quench the reaction mixture; (c) heating the quenched reaction mixture at 90- 100 °C until the volume of reaction mixture has reduced to 5-15 %; (d) cooling the reduced volume of quenched reaction mixture and adding water and base solution to it to maintain pH of 3 followed by addition of solvent to obtain a reaction mixture; and (e) stirring the reaction mixture at 25-30 °C at 200-400 rpm for 14-16 hours to obtain precipitate of bisphosphonate (I); and (f) washing the precipitate with the ethanol to obtain bisphosphonate (I). USE - Process for synthesizing biocompatible bisphosphonate (I) used for treating post-menopausal osteoporosis, glucocorticoid induced osteoporosis, Paget's disease of bone, metastatic bone diseases, hypercalcemia of malignancy. No biological data given. ADVANTAGE - The process is biocompatible, cost-effective, rapid and efficient. The (I) has high purity. DETAILED DESCRIPTION - Synthesizing biocompatible bisphosphonate of formula (I), involves (a) mixing adipic acid ester derivative, phosphorous acid, and a halo phosphorous compound under stirring at 200-400 rpm to obtain a reaction mixture; (b) heating the reaction mixture within an hour at 60-100 °C under N2 atmosphere and refluxing for 14-16 hours followed by addition of cold water to quench the reaction mixture; (c) heating the quenched reaction mixture at 90- 100 °C until the volume of reaction mixture has reduced to 5-15 %; (d) cooling the reduced volume of quenched reaction mixture and adding water and base solution to it to maintain pH of 3 followed by addition of solvent to obtain a reaction mixture; and (e) stirring the reaction mixture at 25-30 °C at 200-400 rpm for 14-16 hours to obtain precipitate of bisphosphonate (I); and (f) washing the precipitate with the ethanol to obtain bisphosphonate (I). |
