成果简介NOVELTY - Carrying out rapid, visual, sensitive, and specific diagnosis of tuberculosis (TB), which involves: (a) setting up an reverse transcriptase loop-mediated isothermal amplification (RT-LAMP) reaction for amplification of Mycobacterium tuberculosis (MTB) nucleic acid RNA using primers specific to insertion sequence (IS) 6110 (IS6110) or MTP40 genes for approximately 40 minutes, where the positive result is indicated by a color change from pink to yellow; (b) forming a nucleoprotein complex by adding Cas enzyme and respective guide RNAs specific to IS6110 or MTP40 genes, and incubating for approximately 30 minutes; (c) setting a trans-cleavage reaction by adding the nucleoprotein complex, RT-LAMP positive product, and a labeled probe, and incubating for approximately 60 minutes; and (d) providing visual detection on a lateral flow dipstick where positive and negative results are interpreted after 3 minutes based on the pattern of control and test bands. USE - Method for carrying out rapid, visual, sensitive, and specific diagnosis of tuberculosis using kit. ADVANTAGE - The method targets the IS6110 and MTP-40 genes of Mycobacterium tuberculosis (MTB) for RNA detection in both bacterial cultures and patient samples, and provide an accurate, quick, and user-friendly TB diagnosis. The use of kit minimizes false positives and false negatives, and enable rapid on-site TB diagnosis with minimal training. DETAILED DESCRIPTION - An INDEPENDENT CLAIM is included for kit for diagnosing TB, which comprises: an RT-LAMP amplification system including primers specific to IS6110 or MTP40 genes and a colorimetric master mix; clustered regularly interspaced short palindromic repeats (CRISPR) system including a Cas enzyme, guide RNAs specific to IS6110 or MTP40 genes, a labelled single-stranded DNA probe, NEB buffer, and nuclease-free water; and lateral flow dipstick for visual readout.
