| 成果简介 | NOVELTY - Preparing copper-coumarin electrodes involves (a) creating a copper compound ink by adding the copper-coumarin compound (I) to polar solvent and buffer and placed in ultrasonic bath for 2-5 minutes, (b) entrapping the solution in a polymer based matrix, (c) mixture blended well and placed on ultrasonic bath for further 2-5 minutes, (d) blending mixture drop cast (50-200 µL) onto metallic disc electrode and dried in air for 30-60 minutes The resultant copper-coumarin electrode is then washed carefully with water and PBS (Phosphate-buffered saline) for three times each to obtained copper-coumarin electrodes as electrochemical sensor. USE - Method for preparing copper-coumarin electrodes for electrochemical sensing of homocysteine (Hcy). ADVANTAGE - The copper-coumarin electrodes based electrochemical sensor can be measure Hcy selectivity in serum sample even at a low concentration (1 µM). The copper-coumarin electrodes based electrochemical sensor is suitable for measuring Hcy in real time analysis since no requirement of binding time and the electrode modification procedure is simple. The limit of detection and limit of quantification for Hcy is 0.025 µM and 0.077 µM with R2 square value 0.96. The peak current observed at the peak potential showed a linear increase in current with respect to Hcy concentrations. Other biothiols such as cysteine (Cys) and glutathione (GSH) did not produce any detectable signal at the peak potential observed for Hcy, indicating the selective nature of the electrosensor towards Hcy. There is no requirement of binding time for the proposed sensor which is essential for real time analysis and point-of-care diagnosis. The life-time of the copper coumarin electrodes is more than one year which is comparable to other affinity based sensing platforms involving antibodies. The analytical tool involves a simple electrode fabrication process (4-5 mins) making it much easier and less time consuming. The linearity range obtained (5 to 100 µM) for measuring Hcy in real human serum makes it suitable for detecting analysing clinical conditions such as hyperhomocysteinemia.
