USE - Method for preparing omidenepag isopropyl compound used in preparation of medicament for treating elevated intraocular pressure in patients (all claimed). No biological data given. ADVANTAGE - The compound (I) has purity of more than 99% by HPLC, and is simple at a large scale production with economically significance. DETAILED DESCRIPTION - Preparing omidenepag isopropyl compound of formula (I) comprises: reacting N-{[4-(1H-pyrazol-1-yl)phenyl]methyl}-N-[(pyridin-2-yl)methyl]pyridine-3-sulfonamide compound of formula (III) with a acetate compound of formula (II) in the presence of a base and a solvent to obtain a compound (I). R1=an amino protecting group, preferably 9-Fluorenylmethyloxycarbonylation (Fmoc), tertButoxycarbonylation (Boc) reagents, Allyloxycarbonylation (Alloc) reagents, Benzyloxycarbonylation (Cbz) reagents, Benzylation (Bn) reagents, Allylation (All) reagents and 2,2,2-Trichloroethoxycarbonylation (Troc) reagents; and X=halogen group selected from iodo, chloro, fluoro and bromo groups. INDEPENDENT CLAIMS are included for: 1. a process for preparation of compound of formula (IV) comprising: reacting compound (VI) with compound (V) in the presence of a base and a solvent to form a compound (IV); and 2. a composition of omidenepag isopropyl comprising the impurity-A, of less than 0.1% by HPLC. |
