成果简介NOVELTY - Composition of DNA loaded alpha lactalbumin nanoparticles is prepared by: (a) preparing GFP-p53 DNA-loaded lactoferrin and alpha lactalbumin nanoparticle by preparing Lactoferrin nanoparticles using the sol-oil method, taking 1:10 molar ratio of DNA: Alpha lactalbumin and incubating for overnight at 4℃; sonicating mixture with amplitude of 40 and 30 seconds on and off pulse for 5 minutes, on ice; repeating above pattern for three times, immediately snap frozen in liquid nitrogen for 10 minutes and later on thawed on ice, pelleting down nanoparticles down at 6000 rpm for 10 minutes at 4℃ and washing with ether till the removal of oil traces, re-suspending nanoparticles in 0.1 M phosphate-buffered saline (PBS) buffer pH-7.4, in case of drug, incubating drug for period of saturation as per spectrophotometric analysis; and (b) preparing nanoparticle composition with Lactoferrin and Lactalbumin. USE - Composition of DNA loaded alpha lactalbumin nanoparticles for treatment of virus infection such as HIV, and cardiomyopathy, tissue specific cancers. Test details are described but no results given. ADVANTAGE - The composition reduces such toxicities and provides target direction of natural site of action, thus enhances efficacy; serves a mimic for target localization of API in the heart; ensures stability of therapeutic molecules in the system (in-vivo) and efficacy at site of action. DETAILED DESCRIPTION - Composition of DNA loaded alpha lactalbumin nanoparticles is prepared by: (a) preparing GFP-p53 DNA-loaded lactoferrin and alpha lactalbumin nanoparticle by preparing Lactoferrin nanoparticles using the sol-oil method, taking 1:10 molar ratio of DNA: Alpha lactalbumin and incubating for overnight at 4℃; sonicating mixture with amplitude of 40 and 30 seconds on and off pulse for 5 minutes, on ice; repeating above pattern for three times, immediately snap frozen in liquid nitrogen for 10 minutes and later on thawed on ice, pelleting down nanoparticles down at 6000 rpm for 10 minutes at 4℃ and washing with ether till the removal of oil traces, re-suspending nanoparticles in 0.1 M phosphate-buffered saline (PBS) buffer pH-7.4, in case of drug, incubating drug for period of saturation as per spectrophotometric analysis; and (b) preparing nanoparticle composition with Lactoferrin and Lactalbumin at ratio of 80:20 and 60:40 by dissolving weighed 1 mg rhodamine 6G in water and lactoferrin and lactalbumin in the ratio of 80:20 and 60:40% (i.e., 3.2 mg and 0.8 mg and 2.4mg and 1.6 mg respectively) and dissolving in 1X PBS(pH 7.4), mixing and incubating on ice for 1 hour; after 1 hour, adding 2 ml olive oil while vortexing slowly, sonicating for 15 minutes at 40 Hz amplitude with 5 minutes time interval; snap freezing in liquid nitrogen for 10 minutes, thawing on ice for 3 hours, centrifuging at 6000 rpm for 10 minutes, removing supernatant and washing the pellet with diethyl ether, and dissolving airdried pellets in 1X PBS (pH 7.4).
