成果简介NOVELTY - Recombinant proline-proline-glutamate (rPPE2) protein is new, where rPPE2 protein comprising 561 amino acid sequence of (SEQ ID NO: 2), given in the specification. USE - Recombinant proline-proline-glutamate (rPPE2) protein used as a subunit vaccine for reducing Tuberculosis (TB)-associated adipose tissue dysfunction, insulin resistance and diabetic complications and ameliorates of TB-associated diabetes (claimed). No biological data given. ADVANTAGE - The rPPE2 protein exhibits adipose tissue atrophy along with reduction of adipogenic gene expression, impaired glucose tolerance, and reduction of adipogenic gene expression, shows significant improvement in adipose tissue mass as well as tissue histology indicating the neutralization of a PPE2 protein by an antibody developed through the rPPE2 protein vaccination rescuing the effect of the PPE2 in induction of TB-associated adipose tissue dysfunction and impaired glucose tolerance. DETAILED DESCRIPTION - An INDEPENDENT CLAIM is included for method for development of recombinant proline-proline-glutamate (rPPE2) protein, which involves: (1) culturing of a bacterial strain by generating Mycobacterium smegmatis expressing a PPE2 protein of Mycobacterium tuberculosis as a Msmeg-PPE2, and generating a CDC1551 strain complemented with a wild-type PPE2 as CDC1551:ΔDPPE2::PPE2 by electroporating a pVV16-PPE2 construct into CDC1551:ΔDPPE2; (2) infecting a mice with the bacterial strain by infecting mice with the CDC1551:ΔDPPE2::PPE2 and optionally with CDC1551:ΔDPPE2 via aerosol route to form impurified rPPE2 protein; and (3) purifying the impurified rPPE2 protein to form a rPPE2 protein.
