Test details are described but no results given. ADVANTAGE - The anionic hydrogel system ensures antibacterial and anti-biofilm effectivity, undergoes conformational changes and self-assemble upon variations in pH and/or temperature, and exhibits gelation properties under physiological settings, ensures direct encapsulation of cells in a three-dimensional thixotropic (injectable) matrix. DETAILED DESCRIPTION - A cytocompatible and antibacterial anionic peptide comprising a selective anionic dodecapeptide PEP-M with NH2-Trp-Tyr-Thr-Gly-Glu-Gly-Met-Asp-Glu-Met-Glu-Phe-CONH2(SEQ ID NO: 1) is new, where the peptide has a net charge of -3 at physiological pH of 7.4 constituting a selective αβ-tubulin heterodimer interface region having self-assembly based hydrogel formability. INDEPENDENT CLAIMS are included for: an anionic hydrogel system, which comprises gelled cytocompatible anionic dodecapeptide PEP-M; a method for producing an anionic dodecapeptide PEP-M involving solid-phase peptide synthesis (SPPS); and a method for synthesizing hydrogel, which comprises: (i) dissolving at least 1.8 mg PEP-M selectively in 1 mL filtered 1x phosphate buffer saline in a vial to obtain 1.8 wt. % solution or HPLC purified water; (ii) subjecting the resulting mixture to sonication at 20-40 kHz for 15 minutes to obtain a homogeneous mixture; (c) sonicating gently for 15 minutes at 37℃ temperature water bath or heating at 80℃ followed by cooling at room temperature for 30 minutes; (d) subjecting obtained mixture to rapid supramolecular self-assembly at minimum gelation concentration (MGC) og 1.8 wt.% at physiological pH (pH of 7.4) upon gentle sonication for 15 minutes at 37℃ water bath or heating at 80℃ followed by cooling at room temperature for 30 minutes to obtain hydrogel. |
