成果简介NOVELTY - Invention relates to nursery, to methods of assessing genetic stability of microclonally propagated strawberries. Disclosed is a method for assessing the genetic stability of microclonally propagated strawberry plants, involving sampling a fresh biomaterial, DNA extraction by CTAB and comparative PCR analysis, characterized by the fact that high-information ISSR-marker ASSR 20 or high-information ISSR-markers are used to assess genetic stability of regenerants and mother plant of garden strawberry: UBC 849 and ISSR 6. In this method, the following parameters are used during PCR analysis: 3 minutes of preliminary denaturation at temperature of 95 °C; next 35 cycles: denaturation 35 seconds at 95 °C, primer annealing: 1 minute at 50 °C, elongation: 1.5 minutes at 72 °C, and the final synthesis cycle at temperature of 72 °C for 5 minutes. Method is suitable for implementation in laboratories equipped with a minimum amount of equipment for molecular genetic research. USE - Plant growing.
