| 成果简介 | NOVELTY - Invention relates to a method for editing DNA of the African swine fever virus in MGF 110_4L-6L genes using plasmids U6, pJET1.2 and Cas9 enzyme. Said method provides for the creation of a plasmid-guide (gRNA) based on the plasmid U6, containing 20 bps, complementary to the portion of the genome of the ASF virus with genes MGF 110_4L-6L in positions 8724-10107 nucleotide base of the genome of the ASF virus, creation of shuttle plasmid Shuttle plasmid based on plasmid pJET1.2, which contains two fragments of ASF virus genome—Left-arm (L-arm) and Right-arm (R-arm), flanking MGF 110_4L-6L genes for their deletion, ASF virus genome editing in MGF 110_4L-6L genes region and the selected ASF virus strain in the primary porcine spleen cell culture, extraction of ASF virus DNA, sequencing and confirmation of MGF 110_4L-6L gene deletion.
