成果简介NOVELTY - Invention relates to molecular biology, forensic medicine and forensic science and concerns an instrument for determining sex, assessing the degree of degradation of nuclear and mitochondrial DNA and the presence of PCR inhibitors. According to the results of the fragment analysis, based on the known expected lengths specific for 4 in the case of female DNA or 7 in the case of male DNA fragments, analyzed human DNA and 1 internal positive control (IPC), by the presence/absence of alleledrop-out peaks, their height and area, the DNA sex of the analyzed samples can be primarily assessed by AmelX/AmelY and SRY gene fragments, containing various degrees of degraded genetic material, namely nuclear DNA—by two AmelX/AmelY gene fragments and mitochondrial DNA—by fragment of non-coding region HV1 and ND1 gene and PCR inhibitors—by a fragment of the CSN3 gene of B. tauris, which is IPC. As a result, three parameters for a specific DNA preparation are known per one reaction. Taking into account the results obtained using the described method, at the subsequent stages of the work, the researcher is able to avoid unjustified waste of reagents and time. USE - Genetics.
