| 成果简介 | NOVELTY - Invention refers to medicine, namely to obstetrics and gynaecology, and concerns cervical examination. Uterine cervix is examined in mirrors during colposcopy, cytological examination with verification of diagnosis, PCR examination of cervical epithelium scraping for HPV. Then performing targeted monomultifocal biopsy with primary detection of H-SIL; or primary surgical biopsy in L-SIL, benign hyperplastic processes, recurrent ASC-US. Thereafter, the obtained material is subjected to histological and immunohistochemical analysis using antibodies to p16INK4a and KI-67 proteins to assess the degree of manifestation of proliferative activity, as well as cell atypia. Homogenates are obtained from the obtained biopsy or surgical material by separating a fragment with a diameter of at least 5 mm with the inclusion of an endocervical zone, which is placed in a sterile test tube with addition of 5 ml of physiological solution. Then, during first 2-4 hours from the preparation of the material in the same test tube, it is homogenised with a rotary homogeniser to a homogeneous mass suspended in physiological solution, followed by centrifugation for 15 minutes at 3,000 rpm, then the supernatant in volume of 1 ml is placed into an Eppendorph test tube with a transport medium for collecting and transporting scrapings or smears of epithelial cells, then cervical tissue homogenates with predominance of glandular component are examined by PCR with determination of HPV 14 oncoserotypes with determination of viral load.
