| 成果简介 | NOVELTY - Synthesized/engineered peptide is new, where synthesized/engineered peptide comprises 11 amino acid sequence (SEQ ID NO: 1), given in the specification, and has binding affinity less than -7 kcal/mol. USE - Synthesized/engineered peptide used in manufacturing engineered exosome system and as a stable nano formulation suspended in phosphate buffer saline (claimed) for targeting neural stem cells (NSC). ADVANTAGE - The synthesized/ engineered peptide has molecular weights of 1500.78 Da at 100% purity and 1859.81 Da at 99.72% purity for 5-FAM peptide and is completely non-toxic in concentration levels of as high as 400 μM. The nano formulation comprising surface functionalized select exosome with the synthesized/engineered peptide is adapted to specifically target NSC in adult brain to increase the uptake of exosome by cells to enable therapeutic effect by enhancing NSC proliferation and subsequently differentiation to new neurons, free of immunogenicity and devoid of any cell death by overcoming the problems of cell-based transplantation. The engineered exosome system has the characteristics of saucer shaped morphology (TEM images), enhanced exosome size od 195.76 ± 8.37 nm after peptide functionalization, and it is non-toxic in concentration levels of as high as 80 μg/ml, favours 2-fold increase in cell proliferation as seen in NSC culture post treatment with the engineered exosome (Exo-pep-11) for 72 hours. The peptide in nano formulation specifically targets NSC in adult brain and increase the uptake of exosome by cells, and the exosome shows its therapeutic effect by enhancing NSC proliferation and subsequently differentiation to new neurons. The exosome system formulation being cell derived product is advantageous over the existing solutions for having target specificity, no immunogenicity, avoiding cell death and overcoming the problems of cell-based transplantation. DETAILED DESCRIPTION - INDEPENDENT CLAIMS are included for: Engineered exosome system for NSC targeting engineered exosome, which comprises surface functionalized exosome which is surface functionalized with peptide with SEQ ID NO: 1 and provide for a mutually co-acting NSC targeting therapeutic efficacy; Method for manufacturing the synthesized/engineered peptide; and Method for manufacturing the engineered exosome system, which involves: (1) providing the synthesized/ engineered peptide; (2) providing the cell derived exosome; (3) coupling the carboxy acid group present in the synthesized/engineered peptide with surface amine groups on the exosome protein by EDC/sulfo-NHS in MES [(2-(N-morpholino)ethanesulfonic acid] buffer on a shaker resulting in amide bond formation between the exosome and peptide and obtaining the engineered exosome (Exo-pep-11) and nano formulations. |
