| 成果简介 | NOVELTY - Invention relates to nanotechnology, to methods of producing metal nanoparticles and can be used in medicine, chemical industry, micro- and nanoelectronics, instrument making. Invention is a method of producing a solution of nanoparticles using metal ions Co(II) and Ni(II) inside protein cavity of recombinant bacterial protein Dps Escherichia coli, including hydrolysis in mild conditions of recombinant protein Dps Escherichia coli and simultaneous removal of residues of inorganic components from the protein structure by dialysis with successive maintenance in external buffer solutions with pH = 8.0; 7.0; 6.0; 5.0 and 3.8, taken in 400-fold volume excess relative to the volume of the protein solution, for 60 minutes, in each, at a temperature of +4 °C, in a dialysis bag with length of 5 cm and an inner diameter of 1 cm, with pore size that allows particles with mass of not more than 3.5 kDa, with an aqueous solution containing 1 mg/ml of recombinant protein Dps and 50 mM Tris-HCl with pH 8.0, 0.1 mM EDTA; recovery of the dodecamer structure Dps by subsequent dialysis of the obtained solution of hydrolysed and purified recombinant protein Dps with successive maintenance in external buffer solutions with pH = 3.8; 5.0; 6.0; 7.0; and 8.0; taken in 400-fold volume excess relative to the initial volume of the protein solution, for 60 minutes, in each, at a temperature of +4 °C, in a dialysis bag with length of 5 cm and an inner diameter of 1 cm, with a pore size that allows particles with a mass of not more than 3.5 kDa, with an aqueous solution of recombinant hydrolysed and purified protein Dps; adding a source of transition valence metal ions; oxidation of transition valence metal ions absorbed by the recombinant bacterial apo-protein Dps, air oxygen dissolved in the buffer solution under normal conditions, while incubating the obtained mixture for 60 minutes at room temperature; removal of unbound metal ions by dialysis while holding for 60 minutes at a temperature of +4 °C of dialysis bag 5 cm long and an inner diameter of 1 cm, with pore sizes that allow particles with a mass of not more than 3.5 kDa, with a solution of recombinant protein Dps, saturated with metal ions, in 400-fold volume excess relative to the initial volume of the protein solution, an external solution containing 50 mM Tris-HCl with pH 8.0, 0.1 mM EDTA, 50 mM NaCl, according to the invention, the source of metal ions with transition valence is an aqueous solution of CoSO4 with a volume ratio of 1 mg/ml protein solution: 150 mM CoSO4 solution equal to 1:2.5, or aqueous solution of NiSO4 at volume ratio of 1 mg/ml protein solution: 100 mM NiSO4 solution equal to 1:2.5. USE - Chemistry; biotechnology.
