成果简介NOVELTY - Abiotic paper-based colorimetric test kit/system comprises: (a) methylene blue dye and ferrous sulfate (FeSO4)-coated paper substrate-based colorimetric change-based detector; and (b) oxidant including hydrogen peroxide (H2O2). The methylene blue dye and FeSO4coated paper substrate based colorimetric change based detector is adapted for the H2O2solution treated testing of samples for indicating the bacterial detection with readily noticeable colorimetry based color change in the methylene blue dye paper strip and co-acting divalent cations (Fe2+), where the absence of any bacterial contamination is indicated by ready de-coloration of the methylene blue dye paper strip and the presence of bacterial contamination is indicated by its related readily quantifiable level of bacterial contamination based color retention on it for onsite detection. USE - Abiotic paper-based colorimetric test kit/system for onsite bacterial detection in samples including catalase-positive bacteria. ADVANTAGE - The abiotic paper-based colorimetric test kit/system has a limit of detection of 20 colony forming units (CFU)/ml, and shelf life of at least 6 months when prevented from light exposure, measures catalase activity of contaminating bacteria in water with a response time of 5-10 minutes, and excludes pre-culturing/enrichment/enumeration of the sample and any instrumentation to read the results, and is sensitive, user-friendly, and cost-effective for efficient and rapid testing of bacteria including coliforms, provides a scalable, economical, and portable bacterial detection method, and increases the shelf life and portability using abiotic components in the sensor. DETAILED DESCRIPTION - INDEPENDENT CLAIMS are included for: 1. a method for manufacturing the abiotic paper based colorimetric test kit/system comprising: (i) providing methylene blue dye and FeSO4coated paper substrate-based colorimetric change-based detector including coating a filter paper substrate with a mixture of a dye and divalent cation, selected from methylene blue and FeSO4; and drying the dye-coated paper at room temperature around 30± 5℃; (ii) providing 35 mM H2O2solution; and (iii) detecting the changes in strip color and its related quantification by naked eye; and 2. a method for detecting bacteria from test samples including water comprising: (i) treating the methylene blue dye and FeSO4-coated paper substrate with H2O2; (ii) dipping the H2O2treated methylene blue dye and FeSO4coated paper substrate in the sample to be tested; and (iii) observing colorimetry-based color change in methylene blue dye paper strip and co-acting divalent cations (Fe2+).
