| 成果简介 | NOVELTY - Micropropagation system (100) comprises a surface sterilization chamber (102) including one or more ultraviolet ray lamps to sterilize strawberry runner tip explants, a sterile culture generation chamber (104) to culture the sterilized explants in a controlled culture room, where temperature is maintained between 22℃ to 23℃ and a photoperiod of 12 hours a day. A multiple shoot regeneration chamber (106) is set for inducing shoot regeneration in the explants, a root initiation chamber (108) is set for inducing root formation in the regenerated shoots, and an acclimatization chamber is used for acclimatizing the rooted plantlets to ex vitro conditions. USE - Micropropagation system for enhanced strawberry plant multiplication and tissue culture. ADVANTAGE - The micropropagation system ensure uniform light exposure and nutrient absorption from all directions, and promote even and accelerated strawberry plant shoot development. DETAILED DESCRIPTION - An INDEPENDENT CLAIM is included for method of micropropagation, involves: (1) carrying out surface sterilizing runner shoot tips of strawberry plants; (2) preparing sterile culture medium comprising modified Murashige and Skoog (MS) basal medium, 3 wt.% sucrose, and 0.8 wt.% agar; (3) culturing the sterilized runner shoot tips obtained in step (1) in the sterile culture mediumprepared in step (2) in a controlled tissue culture room maintained at a temperature of 22℃-23℃ with a photoperiod of 12 hours a day for 1 week; (4) preparing a multiple shoot regenerating medium comprising modified MS basal medium, 3 wt.% sucrose, 0.8 wt.% agar, and 2 mg/L metatopollin growth hormone; (5) culturing and subculturing the sterilized runner shoot tips obtained in step (3) in the multiple shoots regenerating medium prepared in step (4) an initial inoculation period of 2 weeks, then subculturing after 1 week in the same medium, and conducting in a controlled tissue culture room at a temperature of 22℃-23℃ with a photoperiod of 12 hours a day; (6) preparing a rooting medium comprising 1/2 MS basal medium, 3 wt.% sucrose, and 0.6 wt.% agar; and (7) transferring the growing shoots obtained in step (5) into the rooting medium prepared in step (6) by placing the growing shoots into the rooting medium for 3 weeks in a controlled room at a temperature of 22℃-23℃ with a photoperiod of 12 hours a day, and achieving a root regeneration frequency of 95-99%.
