| 成果简介 | NOVELTY - Stable extracellular fungal laccase comprises benzenediol and oxidoreductase, and it is produced by white-rot fungi Ganoderma multipileum, bearing the International Enzyme Commission number E.C. 1.10.3.2., and carrying out biobleaching without the use of mediators. USE - Stable extracellular fungal laccase used as biobleaching agent in textile industry. ADVANTAGE - The stable extracellular fungal laccase is stable in the range of 30-80℃, shows increase in activity, in the presence of 10-20 vol.% organic solvents including ethanol, methanol, acetonitrile and benzene. The enzyme retain 75% and 55% of initial activity at 0.1M and 0.5M sodium chloride, and its stability more than 50% for 24 hours in the presence of metal ions, namely ferrous (iron(II)) ion (Fe2+), potassium ion (K+), zinc ion (Zn2+), nagnesium ion (Mg2+), sodium ion (Na+), and calcium ion (Ca2+). The kinetic properties of laccase is Km value is52.66mM, Vmax value is 9.27μM/ml/min and Kcat value is 463.85. The laccase avoid the use of harmful bleaching agents and chemicals which damages the fiber of the fabric. The white-rot fungi are the only microorganisms which are capable to degrade the entire wood components. The biobleaching process of cotton fabric by laccase eliminate the use of mediators, and it simplify the process of bleaching, reduce additional chemical inputs and minimize by-product formation. DETAILED DESCRIPTION - An INDEPENDENT CLAIM is included for method of preparing stable extracellular fungal laccase, which involves: (1) soaking desized and scoured fabric in water in 1:30 material liquid ratio; (2) adding accase at pH 6 to the soaked fabric obtained in the concentration 1ml/l to obtain a reaction mix and adding 2g/l Triton (RTM: nonionic surfactant) X-100 to the reaction mix obtained; (3) incubating the samples at 50℃ in an incubator shaker bath at stirring speed of 100 revolution/minute (rpm) for 1 hour; and (5) washing the samples thoroughly and drying. |
