成果简介NOVELTY - Disclosed is a method for assessing neurotropic and/or neurotoxic activity of substances and compositions thereof. Method involves separation of spinal ganglia from neonatal piglets aged up to 3 days; obtaining a monolayer culture from them in an alpha-MEM medium with 10% foetal calf blood serum and with the addition of an antibiotic and antimycotic, reseeding in concentration of 2.5-5×103 cells/ml on 8-10th day of growth; or obtaining multicellular spheroids formed by cells recovered from spinal ganglia of neonatal piglets aged up to 3 days, in alpha-MEM medium with blood serum substitutes B27, Neuromax and adding an antibiotic and an antimycotic; introduction of multiple concentrations of the test samples into the wells with the obtained culture of not more than 5th passage or onto the microelectrode systems on which the culture of not more than 5th passage is placed, or in wells with multicellular spheroids in concentration of not less than 10 spheroids with diameter of 100-200 mcm per well, as well as control samples with known neutrotropic or neurotoxic action; evaluation of parameters of proliferative activity of culture cells, visual cytotoxic effects and/or electrophysiological activity. Result is evaluated by comparison with reference samples and/or by calculating ED50 and/or LD50 by probit analysis. USE - Biotechnology.
