成果简介NOVELTY - Hexavalent combination vaccine comprises a combination of polysaccharidecarrier protein glycoconjugates of serogroups A, C, Y, W, and X of Neisseria meningitidis, and one or more proteins of serogroup B of Neisseria meningitidis. USE - Hexavalent combination vaccine used in pharmaceutical composition for treatment, prevention or diagnosis of meningitis (claimed). No biological data given. ADVANTAGE - The hexavalent combination vaccine include adjuvants, which shows enhancement of the immunogenicity of antigens, modification of the nature of the immune response, reduction of the antigen amount needed for a successful immunization, frequency of booster immunizations needed and improved immune response in elderly people. The pharmaceutical composition has a long shelf-life of 12 to 36 months. The vaccine is capable of complete prophylaxis against the meningococcal diseases, and the composition fulfils the unmet medical need for Meningitis prophylaxis and treatment. DETAILED DESCRIPTION - INDEPENDENT CLAIMS are included for: Method of preparing the polysaccharide-carrier protein glycoconjugates of serogroups A, C, Y, W, and X of Neisseria meningitidis, which involves: (1) cultivating meningitis serogroup strains A, C, Y, W and X in presence of complex media for about 12-20 hours to produce meningococcal A, C, Y, W, and X polysaccharides; (2) purifying the meningococcal A, C, Y, W, and X polysaccharides by processes selected from ultrafiltration, cetyl-trimethylammonium bromide (CTAB) precipitation, centrifugation, depth filtration, ethanol, and salt precipitation; (3) activating each of the purified meningococcal A, C, Y, W, and X polysaccharides at an alkaline pH of 8.5-9.5 in the presence of 1-Cyano-4 Dimethylaminopyridine Tetrafluoroborate (CDAP) and at a temperature ranging from 8-15?oC; (4) activating or derivatizing the carrier protein by 1-ethyl-3-(dimethylaminopropyl)- carbodiimide (EDAC) for about 16-24 hours; (5) mixing and conjugating the activated A, C, Y, W, and X polysaccharides and the activated carrier proteins to obtain the polysaccharide-carrier protein glycoconjugates of serogroups A, C, Y, W, and X of Neisseria meningitidis; and Pharmaceutical composition comprises, hexavalent combination vaccine, a pharmaceutically acceptable carrier, 2μg-10μg per dose polysaccharide-carrier protein glycoconjugates of serogroup A of Neisseria meningitidis; 2μg-10μg per dose polysaccharide-carrier protein glycoconjugates of serogroup C of Neisseria meningitidis, 2μg-10μg per dose polysaccharide-carrier protein glycoconjugates of serogroup Y of Neisseria meningitidis, 2μg-10μg per dose polysaccharide-carrier protein glycoconjugates of serogroup W of Neisseria meningitidis, 2μg-10μg per dose polysaccharide-carrier protein glycoconjugates of serogroup X of Neisseria meningitidis, 60 μg-240 μg per dose proteins of serogroup B of Neisseria meningitidis, 150 μg- 500 μg per dose diluent or adjuvant, 10-30 mg per dose sucrose, 0.01-0.04 ug per dose emulsifier, 1μl to 4 μl per dose preservative, and buffer to make the pH 6.8 ± 0.4.
