成果简介NOVELTY - Proposed method for selection of probiotic and autoprobiotic strains of microorganisms by resistance to cytostatics in therapy of colorectal cancer, consisting in the fact that inoculums of pure cultures of probiotic strains of enterococci and lactobacilli and similar taxa of autoprobiotic strains isolated from faeces of patients with colorectal cancer, grown on MRS agar, diluted in isotonic sodium chloride solution to concentration of 8 lgCFU/ml, then MRS broth is successively introduced into each well of a flat-bottomed microculturing plate, separately tenfold dilutions of cytostatics: fluorouracil, oxaliplatin and leucovorin, and a suspension of pure cultures in amount of 5% of the total volume of the medium, cultivated for 24 hours at 37 °C and measuring optical density of samples at wavelength of 630 nm, compared with the optical density of samples which do not contain cytostatics, determining for each cytostatic the minimum inhibitory concentration which changed the optical density of the medium during the growth of indigenous or probiotic pure cultures by not more than 10%, and selecting for therapeutic purposes bacterial cultures showing maximum resistance to all cytostatics.
