| 成果简介 | NOVELTY - Culturing and expanding human dental pulp stem cells (DPSCs) involves: (a) washing pulp tissue in 1x phosphate buffer saline (PBS) and chopping it into pieces in a media containing antibiotics; (b) treating with 0.1 wt.% collagenase and 50 U/ml dispase in a 37℃ water bath for 2 hours with continuous agitation; (c) incubating the cells for 1 hour at 37℃, then sieving through a 70 μm cell strainer, and centrifuging at speed of 1000 revolution/minute (rpm) for 5 minutes; (d) seeding onto 60 mm dishes in Dulbecco's Modified Eagle Medium (DMEM)/F12 containing 20 wt.% fetal bovine serum (FBS), 1.5mM L-glutamine, 2 wt.% antibiotic (100 U/ml penicillin and 10 mg/ml streptomycin), and growth factors; (e) allowing growth for 7 days, incubated at 37℃ with 5 vol.% carbon dioxide (CO2); and (f) changing the medium in every 3-4 days, and maintaining cells up to 3-4 passages within which all experiments are performed. ADVANTAGE - The culture medium used in the method supports the proliferation and maintenance of DPSCs in an optimal environment. |
