| 成果简介 | NOVELTY - Point-of-care blood plasma-based biomarker detection system comprises: (i) a microfluidic strip comprising a surface-modified Polymethyl Methacrylate (PMMA) microfluidic strip including a separation portion including plasma separator portion having Lateral Flow Assay (LFA) based in-situ plasma separator and a sensing portion including Carbon Dot (CD)-tagged secondary antibody probes and surface-immobilized primary antibodies, adapted for forming complexes upon sensing of plasma-based target biomarker in analytes; and (ii) an image acquisition unit comprising a fluorescent image acquisition unit for capturing fluorescent images of tagged analytes and intensity-based determinants of biomarker detection. USE - Point-of-care blood plasma-based biomarker detection system used for protein biomarkers detection in in-situ separated plasma. ADVANTAGE - The point-of-care blood plasma-based biomarker detection system detects protein biomarkers in in-situ separated plasma with sensitivity of limit of detection (LOD) of 0.4 ng/mL and 0.9 ng/ml and high specificity. The system has portable image acquisition unit and an AI framework to enable simultaneous analysis of multiple analytes with low-cost, and resource-efficient methods. DETAILED DESCRIPTION - An INDEPENDENT CLAIM is included for method for manufacturing the point-of-care blood plasma-based biomarker detection system, which involves: (1) providing a microfluidic strip having a surface-modified Polymethyl Methacrylate (PMMA) microfluidic strip including a separation portion including a plasma separator part having Lateral Flow Assay (LFA) based in-situ plasma separator and a sensing portion provided with CD-tagged secondary antibody probes adapted for forming conjugates upon sensing of plasma-based target biomarker in analytes and binding with the primary antibodies forming a sandwich; and (2) providing an image acquisition unit comprising a fluorescent image acquisition unit for capturing fluorescent images of tagged analytes and intensity-based determinants of biomarker detection. |
