| 成果简介 | NOVELTY - Carrying out visualization of bacteriophages on solid media during their isolation involves utilizing a growth-friendly chromogen that reacts with a specific host bacterial enzyme, where enzymatic cleavage of the chromogen results in a distinct and intensified color change, enhancing chromogenic visualization and enabling the observation of bacteriophage plaque morphology, while the enzymatic activity or presence of substrate and reaction products does not interferewith bacteriophage growth or viability on the solid media. USE - Method for carrying out visualization of bacteriophages on solid media using composition and kit. ADVANTAGE - The method uses growth friendly dye to enhance visualization during bacteriophage isolation, purification, and enumeration, and uses bacterial host enzyme to impart distinct colour onto the solid media which enhances the visibility of bacteriophages plaques. The method does not interfere with the proliferation of bacteriophages thus, maintain the integrity of bacteriophages, and substrate being growth-friendly chromogen, and method does not require additional time-consuming process to produce chromogenic visual onto the solid media. DETAILED DESCRIPTION - INDEPENDENT CLAIMS are included for: Composition for enhanced one-step visualization of bacteriophages on solid media, which comprises a sample suspected of containing bacteriophages, a phage storage buffer, host bacteria producing a specific enzyme, and a growth-friendly chromogen specific to the host enzyme, where the composition facilitates contrast development between bacterial growth areas and clear plaque areas indicative of bacterial lysis by bacteriophages, while preserving bacteriophage integrity; and Kit for bacteriophage visualization, which comprises a host bacterium that expresses an enzyme capable of cleaving glycosidic bonds to initiate a chromogenic reaction, a growth-friendly chromogenic substrate containing a glycosidic bond or its derivatives that, upon enzymatic cleavage, produces a distinct yellow color on solid media, a solvent for preparing the chromogenic substrate at a specified concentration, a sample containing bacteriophages capable of infecting the host and proliferating on solid media, a growth medium for bacterial culture, and protocol documentation detailing the isolation of bacteriophages including the enzymatic reaction between the host enzyme and chromogenic substrate. |
