成果简介NOVELTY - Carrying out microbial mediated preparation of chitosan from chitin, involves: (a) isolating a chitinolytic bacterium, specifically Streptomyces species RB7AG; (b) inoculating the Streptomyces species RB7AG into a minimal salt medium and colloidal chitin medium to initiate growth and enzymatic activity; (c) incubating the inoculated medium for 72 hours, resulting in the degradation of chitin; (d) centrifuging the culture to separate the supernatant, and collecting the extract for further processing; (e) confirming the formation of chitosan from chitin degradation by Scanning Electron Microscopy (SEM) and Fourier Transformed Infrared Spectroscopy (FTIR) analysis; and (f) lyophilizing the extract to obtain the final bioactive chitosan product. USE - Method for carrying out microbial mediated preparation of chitosan from chitin. ADVANTAGE - The microbial fermentation process results in the utilization of chitin-rich waste with minimal waste generation, contributing to an environmentally sustainable method that avoids harmful by-products, and the process is more economically viable and environmentally friendly compared to traditional chemical extraction methods for chitosan production, due to the absence of harsh chemicals. The chitosan produced exhibit significant antifungal and antibacterial properties, providing effective activity against bacterial species Staphylococcus aureus and Klebsiella, as well as fungal species Aspergillus niger and Fusarium oxysporum, and exhibit biological activity that promotes plant growth and resilience against stress without the use of chemical plant stimulants, making it suitable as a natural bio-stimulant. The chitosan is used as an environment-friendly, edible coating material that delays the rotting and extends the storage life of tomatoes, acting as a protective barrier. The chitosan enhances antioxidant enzyme activity in plants, aiding in the effective scavenging of reactive oxygen species (ROS) generated due to stress, for promoting plant health and mitigating stress effects. The chitosan recovered includes lower molecular weight chitosan, comprising dimers and trimers, which display superior efficiency compared to chemically synthesized alternatives. The biotechnological approach provides a sustainable solution for managing chitin-rich waste and valorizing waste products through the production of bioactive chitosan.
