| 成果简介 | NOVELTY - Isolating DNA from mosquitoes involves: (a) taking 4 adult mosquitoes or 3rd instar larvae and placing them into a 1.5 ml microcentrifuge tube (MCT); (b) homogenizing the sample completely using a micropestle; (c) adding 40 μl 10X Tris-Phosphate ethylenediaminetetraacetic acid (TPE) buffer to the homogenized sample and vortexing the solution for 5 seconds; (d) incubating the solution at 70℃ and stirring speed of 900 revolution/minute (rpm) for 25 minutes using a thermomixer; (e) adding 100 mM sodium hydroxide (NaOH) buffer to the solution and mixing gently by inverting; (f) incubating the solution for 5 minutes at room temperature to facilitate cell lysis and DNA extraction; (g) allowing the cell debris to settle at the bottom of the MCT and collecting the supernatant containing the extracted DNA; and (h) transferring the supernatant to a fresh 1.5 ml microcentrifuge tube (MCT) and storing DNA in nanopure (NF) water at 4℃ for downstream PCR analysis and sequencing. USE - Method for isolating DNA from mosquitoes. ADVANTAGE - The method enables isolation of any mosquito DNA, provide a versatile solution for various mosquito species across diverse environments. The DNA isolation assay is field stable, scalable, and efficient, and provide a practical solution for DNA extraction in resource-limited and rural settings. The method not only simplifies the process of DNA extraction but also ensures high-quality results that are suitable for PCR amplification and species identification, and contribute significantly to the ongoing efforts in mosquito monitoring, vector control, and disease prevention. |
