| 成果简介 | NOVELTY - Carrying out electrochemical detection of dengue NS1 protein in human serum samples by utilizing a three-electrode system comprising a screen-printed carbon electrode (SPCE) as the working electrode, silver/silver chloride (Ag/AgCl) reference electrode, and carbon counter electrode, which involves: (a) modifying the surface of working screen-printed carbon electrode (SPCE) by applying an equimolar mixture of 0.1-2 M l-Ethyl-3-(3-dimethylaminopropyl) carbodiimide (EDC) and 0.1-2 M N-hydroxy succinimide (NHS). USE - Method for carrying out electrochemical detection of dengue NS1 protein in human serum samples by utilizing three-electrode system. ADVANTAGE - The method demonstrates high specificity and sensitivity, with no interference from other bioanalytes, making it a reliable and rapid diagnostic tool for detecting dengue infections. DETAILED DESCRIPTION - Carrying out electrochemical detection of dengue NS1 protein in human serum samples by utilizing a three-electrode system comprising a screen-printed carbon electrode (SPCE) as the working electrode, silver/silver chloride (Ag/AgCl) reference electrode, and carbon counter electrode, which involves: (a) modifying the surface of working screen-printed carbon electrode (SPCE) by applying an equimolar mixture of 0.1-2 M l-Ethyl-3-(3-dimethylaminopropyl) carbodiimide (EDC) and 0.1-2 M N-hydroxy succinimide (NHS); (b) adding 1-5 pL 0.1-10 pg/mL anti-dengue NS1 protein onto the working electrode surface and incubating for 5-40 minutes; (c) washing the working electrode with phosphate-buffered saline (PBS) and air-drying; (d) blocking non-specific binding sites on working electrode by applying 0.05-0.1 wt.% bovine serum albumin (BSA) solution followed by a 0.5-1 wt.% polyvinyl alcohol (PVA) coating and air-diying; and (e) recording peak current density by using differential pulse voltammetry (DPV) for detection of dengue NS1 protein. |
