ADVANTAGE - The nucleotide sequence set facilitates rapid species identification, completed within 24 hours, which significantly reduce the time required for regulatory and enforcement actions, ensure high accuracy in species differentiation and reduce the risk of false positives or negatives. The set is preferably adaptable for large-scale testing scenarios, making it suitable for conservation efforts, legal enforcement, and commercial applications. The method is configured to provide identification or differentiation of the species of air-breathing catfish at juvenile stages including fry, fingerling, and larval stages, and identification of processes samples including fish fillets and cooked fish. INDEPENDENT CLAIMS are included for: DNA-based identification kit, which comprises a nucleotide sequence set, reagents for polymerase chain reaction (PCR) amplification including DNA polymerase, deoxynucleotide triphosphates (dNTPs), and buffer, and instructions for performing a PCR-based identification process; and Method for identifying species of air-breathing catfish, which involves: (1) extracting genomic DNA from a sample; (2) amplifying a specific genomic DNA region shared by Clarias magur and Clarias gariepinus using a nucleotide sequence set to obtain an amplified product; and (3) identifying and differentiating Clarias magur and Clarias gariepinus by examining the amplified product. |
