成果简介NOVELTY - Portable kit comprises an eye drop selected from such as xylocaine (lidocaine), proparacaine, and lignocaine, preferably 0.5% proparacaine; a plural of glass dropper bottles of 2-6 ml capacity, containing working solutions of essential reagents including alcohol, 1 % oxidizing agent, Schiff reagent, hematoxylin solution, xylene dips, and mounting medium; self-standing centrifuge tubes of 50-100 ml capacity having screw caps to aliquot the working solutions of the essential reagents, for performing staining process; sterilized filter paper strip/ block to collect test sample for staining; sterilized 2-5 earbuds for pressing the filter paper strip/block and collecting the test sample; sterilized metallic forceps having blunt and round tip for picking and holding the sterilized filter paper strip/ block after collecting the test sample on it; and a manual guide having instructions for using the kit. USE - Portable kit for sample staining in impression cytology. ADVANTAGE - The kit provides quick examination of cells of interest, specifically ocular cells; and is cost-effective, and time-saving. The method comprises minimal steps that provide complete staining of collected cells sample in just 35 minutes after sampling; and is easy, and concise. DETAILED DESCRIPTION - Portable kit comprises an eye drop selected from such as xylocaine (lidocaine), proparacaine, and lignocaine, preferably 0.5% proparacaine; a plural of glass dropper bottles of 2-6 ml capacity, containing working solutions of essential reagents including alcohol, 1 % oxidizing agent, Schiff reagent, hematoxylin solution, xylene dips, and mounting medium; self-standing centrifuge tubes of 50-100 ml capacity having screw caps to aliquot the working solutions of the essential reagents, for performing staining process; sterilized filter paper strip/ block to collect test sample for staining; sterilized 2-5 earbuds for pressing the filter paper strip/block and collecting the test sample; sterilized metallic forceps having blunt and round tip for picking and holding the sterilized filter paper strip/ block after collecting the test sample on it; sterilized 2-5 glass slides and 5-10 cover slips for mounting the test sample on it after staining, for further visualization; a glass dropper for dispensing desired solution(s) for mounting the stained cells on one glass slide; and a manual guide having instructions for using the kit. An INDEPENDENT CLAIM is included for a method for staining and visualizing stained cells which involves: (a) adding 1-2 drops of eye drop; (b) swiping a sterilized filter paper strip/block gently on anesthetized area under examination to collect cells sample using sterilized earbud; (c) holding the filter paper strip/block which contains the collected cells sample, with sterilized metallic forceps and immersing it in 90-97% alcohol for 5-20 minutes to fix the collected cells sample on the filter paper strip / block; (d) immersing the filter paper strip/block containing the fixed cells sample in 65-80% alcohol for 1-5 minute to rehydrate the fixed cells sample on the filter paper strip/block; (e) immersing the filter paper strip/block containing the rehydrated cells sample in 0.5-2% periodic acid solution for 1-5 minutes for oxidation of glycol groups present in the cells sample to aldehydes; (f) washing the filter paper strip/block containing the oxidized cells sample in tap water for 15-40 seconds to remove traces of periodic acid; (g) adding 1 ml Schiff reagent for 5-15 minutes to the washed filer paper strip / block followed by washing it in running tap water for 45-90 seconds; (h) dipping the Schiff- stained and washed filter paper strip/block in 1 ml Harris hematoxylin solution for 30-60 seconds for staining the nucleus followed by washing in running tap water for 15-50 seconds, to obtain filter paper strip/block containing completely stained cells sample; (i) keeping the filter paper strip/block containing the completely stained cells sample in 90-97% alcohol for 1-5 minutes followed by dipping it in absolute ethanol for 1-5 minutes with stirring in between, to remove excess stain to obtain filter paper strip/block containing dehydrated cells sample; (j) exposing the dehydrated cells sample on filter paper strip/block to 1 ml xylene to make the filter paper strip/block transparent and to obtain optimally stained cells sample for proper evaluation; (k) keeping the filter paper strip/block containing the optimally stained cells sample on a slide followed by evaporating the xylene; (l) adding dibutyl phthalate polystyrene xylene on the filter paper strip/block containing optimally stained cells sample; and (m) gently applying cover slip on the top, to prepare a slide for visualization and analysis of the optimally stained cells initially collected from the anesthetized area under examination.
