The purpose of this project is to search and develop new methods for extracting biologically active compounds from the leaves and berries of cranberries (Vaccinium vitis-idaea L.) growing in the Arctic zone of the Russian Federation, in particular, on the territory of the Kola Peninsula, for further practical use of extracts as biologically active additives and pharmaceuticals of plant origin within the framework of "green chemistry".
Valuable plants of the Arctic zone containing important groups of biologically active substances (BAS) are berry bushes of the heather family (Ericaceae Juss.), in particular cranberries (Vaccinium vitis-idaea L.). This plant contains flavonoids, polyphenolic acids, anthocyanins, procyanidins, stilbens, triterpenoids, which are important for maintaining human health properties such as antioxidant, antitumor, antibiotic, etc.
In accordance with modern trends in "green chemistry", traditional organic solvents that harm the environment are being abandoned. One of the promising directions in the extraction of BAS from plant raw materials is the use of natural deep eutectic solvents (natural deep eutectic solvents, NADES). However, at the moment, there are very few comprehensive studies on the processes of extraction of BAS from plants of the Arctic zone in general, and leaves or berries of cranberries, in particular, with the help of NADES. Data on the biological activity of NADES, their toxicity and consumption rates, and the biological activity of extracts based on them are insufficient to confidently predict their effect on the body and to organize their widespread use as bases for biologically active additives and, moreover, pharmaceuticals.
The scientific novelty of the Project lies in the use of NADES to extract BAS from the studied plant. In the course of the work, it is planned to obtain new data on the chemical composition of the studied plants and determine the optimal extraction parameters, as well as determine the biological activity of the extracts obtained.
Various chemical analysis methods will be used to analyze the compositions of the extracts obtained. Antioxidant activity will be determined using phosphomolybdate and FRAP methods. The ability to inhibit free radicals will be evaluated using DPPH and ABTS tests. Also, the cytotoxicity of extracts will be determined using MTT, antibacterial activity on cultures of gram-positive, gram-negative bacteria.
The relevance of this research topic is due to a number of normative legal acts currently adopted in Russia, including the Strategy for the Development of the Arctic Zone of the Russian Federation for the period up to 2035. The development of industry in the Arctic zone, the infrastructure of the Northern Sea Route and the need to ensure comfortable living and working conditions for the population of the Far North, as well as the focus of the Russian economy on import substitution, determine the relevance of the development of domestic innovative biologically active additives and pharmaceuticals to reduce the impact of adverse environmental conditions, maintain human health, and improve the quality of life. The use of local sources of biologically active substances will reduce transportation costs, as well as serve to develop the economic potential of the North. The results of the research planned within the framework of this Project will become the scientific basis for solving important scientific and technical problems in these areas of development of high-tech technologies.
